Population genetic analysis on 23 Y-STR loci using the PowerPlex? Y23 kit in the Mongolian population

Abstract

We have analyzed twenty three Y-chromosomal short tandem repeats(Y-STRs: DYS19, DYS385 a/b, DYS389I/II, DYS390, DYS391, DYS392, DYS393, DYS437, DYS438, DYS439, DYS448, DYS456, DYS458, DYS635, Y-GATA-H4, DYS481, DYS533, DYS549, DYS570, DYS576 and DYS643) loci using the PowerPlex&#9415; Y23 System in 117 unrelated males from Mongolian populations to establish Mongolian Y-STRs database. A total of 112 different haplotypes were identified, of which 109(93.2%) were unique. The most frequent haplotype surveyed herein was found in four individuals(MN 7; 3.4%). The haplotype diversity and discrimination capacity were found to be 0.9934 and 0.9573, respectively. Based on the result of Y-STRs, although Mongolia is geographically located in Northeastern Asia, pairwise RST genetic distances and multi-dimensional scaling(MDS) plot showed that the Mongolian population was found rather close to Europeans. Pairwise RST genetic distances calculated with the Mongolian minor-populations(Bayard, Dorwod, Khalkh and Others) revealed a genetic homogeneity in the Mongolian populations, except for the Bayard ethnic group compared with Khalkh(p < 0.01). This survey shows that the present data of Mongolian individuals could be helpful to establish a comprehensive forensic reference database for frequency estimation.

Abstract

We have analyzed 23 Y-chromosomal short tandem repeats loci (YSTRs: DYS19, DYS385a/b, DYS389I/II, DYS390, DYS391, DYS392, DYS393, DYS437, DYS438, DYS439, DYS448, DYS456, DYS458, DYS635, Y-GATA-H4, DYS481, DYS533, DYS549, DYS570, DYS576, and DYS643) in 261 males from the Mongolian population using the PowerPlex&#9415; Y23 system. Two hundred and thirty one different haplotypes were identified, among which 214 (92.64%) were unique. The most frequent haplotype surveyed was found in seven individuals (3.03%). The haplotype diversity and discrimination capacity were found to be 0.9982 and 0.8851, respectively, which is high degree of polymorphism. Population comparisons based on results of pairwise RST genetic distances and multi-dimensional scaling (MDS) plot showed that the Mongolian population was found to be rather close to Europeans, despite geographical distance. Thus, the present data on the 23 Y-STRs of Mongolian individuals may help establish a comprehensive reference database for forensic casework.
